Expression of Streptococcus Mutans Glucosyltransferase-B in Streptococcus Gordonii

Objective: To investigate the influences of Streptococcus mutans glucosyltransferase-B (gtfB) on the biochemical characteristics of Streptococcus gordonii. Methods: Escherichia coli-Streptococcus shuttle plasmid pZB1 S. mutans gtfB transformed into S. gordonii. Western blot was used to determine whether gtfB was expressed in the transformant. Colony morphologies and in vitro sucrose-dependent adherence were deteced to investigate the biochemical characters of S. gordonii before and after being transformed. Results: The colony morphologies of S. gordonii became smaller than the wild type. Western blot showed that one protein of the mutant reacting with the antibody against S. mutans gtfB glucan-binding fragment had the same molecular weight as that of S. mutans MT8148 cell-associated glucosyltransferase. In vitro sucrose-dependent adherence of S. gordonii decreased significantly after being transformed. Conclusions: The expression of S. mutans gtfB in S. gordonii increased coaggregation of the cells.
