The Biological Effects of Recombinant Human Bone Morphogenetic Protein 2 on Human Periodontal Ligament Fibroblasts

Objective: To evaluate the biological effects of recombinant human bone morphogenetic protein 2 (rhBMP2) on human periodontal ligament fibroblasts (HPLFs). Methods: Primary cultures of HPLFs were maintained in differing concentrations of rhBMP2. The effects on their proliferation, alkaline phosphatase (ALP) activity, osteocalcin synthesis and formation of the mineralized nodules were assessed, respectively. Results: RhBMP2 (0.25 to 2 mg/mL) had no remarkable effect on the proliferation of HPLFs. The ALP activity, osteocalcin synthesis and formation of mineralized nodules of HPLFs were significantly stimulated by 0.5 to 2 mg/mL rhBMP2. Conclusion: The effects of rhBMP2 on HPLFs are dose-dependent. RhBMP2 can stimulate the expression and promote the maturation of osteoblastic phenotypes of HPLFs.
