The Translocation of Smad1 in Human Dental Papilla Cells Treated with rhBMP2

Objective: To investigate the expression of Smad1 in human dental papilla cells and the transduction process, and to explore the molecular mechanism of odontoblast differentiation during tooth development at signaling transduction level. Material and Methods: Immunohistochemical staining of human dental papilla cells derived from primary culture on stimulation by transforming growth factor beta-1 (TGF-beta1) or recombinant human bone morphogenetic protein 2 (rhBMP2). Results: The expression of Smad1 was found in human dental papilla cells upon stimulation by TGF-beta1 and rhBMP2. However, TGF-beta1 and rhBMP2 have no effect on the expression of Smad1 in human dental papilla cells. Smad1 translocated from cytoplasm to nucleus when cells were treated with rhBMP2 but not with TGF-beta1. Conclusion: The expression and translocation of Smad1 in human dental papilla cells were detected for the first time. The signaling transduction of rhBMP2 in human dental papilla cells could be mediated by Smad1.
