The Effect of PLA Membrane Combined with bBMP-PLA Composite on Repair of Rabbit Mandible—A Preliminary Study

Objective: To evaluate the synergistic effect of the combined application of the bBMP-PLA composite and PLA membrane and the effect of PLA membrane on bone-defect healing in the rabbit mandible. Methods: Bilateral and complete bone defects of standard size were created in the mandibular angles of 12 rabbits. The bone defects in each mandible were then covered on both the medial and lateral aspects of the defects with PLA membrane. The bBMP-PLA composite was interpositioned between 2 PLA membranes on the experimental side and the opposite side served as a control. At 4 weeks and at 12 weeks postsurgery, 6 rabbits were sacrificed and observed by gross observation, radiography, light-microscopy, and bone-densitometer examination. Results: The use of bBMP-PLA composite significantly promoted repair of the defects. After 4 weeks of healing, osteogenic repair was observed with osteoid formation and osteogenic cells clustering around the bBMP-PLA composite. By 12 weeks, the primary trabeculae of woven bone has completely bridged the defects. The value obtained by bone-densitometer on the experimental side was significantly higher than that on the control side (P < .01). Conclusion: the bBMP-PLA composite may activate local osteoinductive behavior and maintain the space under the PLA membrane, which would greatly enhance the effect of the PLA membrane.
