Expression and Significance of Connexin43 in Tongue Squamous Cell Carcinoma

Objective: To investigate the role of connexin43 on the development of human tongue carcinoma. Methods: Immunohistochemistry was performed on paraffin-embedded tissue sections of 6 cases of normal tongue epithelium and 24 cases of different differentiated tongue carcinoma to detect the changes of connexin43 expression in the development of human tongue carcinoma. Image analysis was used to quantify the expression levels. Results: In all normal tongue epithelium, connexin43 proteins were localized in cell membranes in spinous layers. There were no significant differences in the area and grayscale of connexin43-positive spots between normal epithelium and well-differentiated squamous cell carcinoma (P > 0.001), but the area of connexin43-positive spots was significantly reduced as cancer cells became morphologically less differentiated. Furthermore, the expression of connexin43 was aberrant in the cytoplasm in tongue carcinoma. Conclusions: The data presented here suggest that quantitative and qualitative changes in connexin43 expression are associated with progression and differentiation of human tongue carcinoma.
